I have got the same problems. And in my case, there are other stuff on the USB-Ports: Steering Wheel, Mouse, Speakers, Printers.

Just keyboard and mouse. Nothing else!
Hmm, maybe it's because of the Energy-Settings in the Windows System Control. Try to turn off the USB energy saving options, maybe this helps. Or if you have an PS/2 Adpater, try this one out.

It can't be that, because one of the very first things I do after installing Windows on ANY desktop PC is to turn all power saving options OFF!

I have the same problem. Also zooming (in/out) and rotating the camera with the mouse doesn't work anymore. (Even though, the mouse cursor is still visible and moving and the icon changes if I press the right mouse button for instance). I cannot link it to (a combination of) keys pressed. For me, it mostly happens right after I tried to place a new bus, but it (sometimes) also happens while driving.
Hope someone will have a solution for this soon.
p.s. Nothing else is connected to USB-ports, except for keyboard and mouse.

OMSI runs with admin rights? Try also the XP compatibility Mode.
Admin rights, 4GB patch but no compatibility mode.
